â Based on the Modified Ferrozine Colorimetric Method
â Iron bound to transferrin is released in an acidic medium and reduced to the ferrous state
â Ferrous ions react with Ferrozine reagent to form a stable colored complex
â Color intensity is directly proportional to the iron concentration
â UIBC is determined by measuring the unbound iron-binding capacity after the addition of excess iron
